The site command to xray Twitter of course:

site:twitter.com graphic (artist | ~design) -job -jobs “atlanta”

site:twitter.com "bio * motorola" (Chicago OR Illinois)

The following operaters to search directly on Twitter:

http://search.twitter.com/operators and http://search.twitter.com/advanced

Example: sql near:Boston within:25mi

There are many sites/applications such as:

http://www.twellow.com/

I like this one to find candidates that mention a particular skillset in their bio or their username. In this case web or software developers. You can refine this further by adding more specific key words, like Java.
site:twitter.com intitle:"on twitter" (intitle:developer|"bio * developer"|"bio * * developer"|"bio * * * developer"|"bio * * * * developer"|"bio * * * * * developer"|"bio * * * * * * developer"|"bio * * * * * * * developer") (web OR software)

Works great with Diver as well. :)
